ActinBrite™ High Affinity Phalloidin Conjugates

Bright, Stable F-actin Staining for Long-Term Imaging

ActinBrite™ High Affinity Phalloidin Conjugates from Biotium are advanced fluorescent dye conjugates of phalloidin, meticulously engineered to retain high binding affinity for F-actin. These novel reagents deliver bright, specific staining that remains stable and reliable for over a month, enabling delayed imaging without compromising signal intensity or specificity.

Key Features

  • Preserves High Affinity for F-actin: Engineered to maintain strong phalloidin binding for vivid and accurate F-actin visualization.

  • Long-Term Stability: Staining remains bright and specific for over a month or longer, depending on the conjugate and storage conditions.

  • Multiplex Ready: Available in five distinct fluorescent colors—from green to near-infrared—ideal for seamless multiplexing.

  • Direct Replacement: Compatible with all standard phalloidin conjugates for straightforward integration into existing protocols.

  • No Signal Fading: Overcomes the limitations of conventional phalloidin conjugates, which typically degrade within a week in standard mounting media.

Fluorescence comparison of ActinBrite™ 488/505 and 665/690 conjugates versus competitor phalloidins over 15 days in fixed HeLa cells, highlighting long-term signal retention of ActinBrite™ dyes.
PFA-fixed, Triton® X-100 permeabilized HeLa cells were stained with ActinBrite™ 488/505 High Affinity Phalloidin, ActinBrite™ 665/690 High Affinity Phalloidin, or the leading competitor dye phalloidins with spectrally similar excitation and emission. Cells were imaged in EverBrite™ Mounting Medium, then stored at 4°C, protected from light, and imaged again using the same microscope settings after 15 days.
Comparison of ActinBrite™ 610/630 and Rhodamine Phalloidin staining in HeLa cells at 3 days and 2 weeks post-fixation. ActinBrite™ shows superior long-term fluorescence signal stability
PFA-fixed, Triton® X-100 permeabilized HeLa cells were stained with ActinBrite™ 610/630 High Affinity Phalloidin or Rhodamine Phalloidin. Cells were mounted in EverBrite™ Mounting Medium and stored at 4°C, protected from light, and then imaged after 3 days or two weeks using the same microscope settings.

Traditional fluorescent phalloidins can suffer reduced binding affinity due to dye conjugation, often leading to diminished signal and short-term stability. ActinBrite™ solves this challenge with a novel design that ensures enduring, high-fidelity F-actin staining. Whether used in fixed cells or tissue samples, ActinBrite™ provides dependable performance even in delayed imaging workflows.

These conjugates are part of Biotium’s broader range of high-performance fluorescent tools, including cellular stains, viability probes, and biomolecule conjugates using their proprietary CF® Dyes—known for exceptional brightness and photostability.
