ExoBrite™ True EV Membrane Stains are advanced lipophilic dyes engineered specifically for extracellular vesicle (EV) research. Unlike conventional membrane dyes such as PKH, DiO, and DiI, ExoBrite™ stains deliver near-complete EV coverage with minimal aggregation, ensuring accurate EV detection across diverse experimental workflows.

These dyes are validated on leading EV analysis platforms, including Particle Metrix ZetaView® and Spectradyne ARC™ Particle Analyzer, making them the gold standard for fluorescence nanoparticle tracking analysis (fNTA) and flow cytometry-based EV detection.

Proven Results: Significantly Greater Coverage of EVs Over Other Membrane Dyes Such as PKH, DiO, and DiI

SEC purified MCF-7-derived EVs were stained with three green membrane dyes: ExoBrite™ 515/540 True EV Membrane Stain, PKH67, and Neuro-DiO. The stained EVs were diluted and run on a ZetaView® particle analyzer. Total EV number was measured in scatter mode, and fluorescently-stained EV number was measured with 488 nm excitation and 500 nm filter. The percentage of particles stained with the dye is shown in the plot. ExoBrite™ 515/540 True EV Membrane Stain showed nearly complete coverage of EVs, much higher than PKH67 or Neuro-DiO.

SEC purified MCF-7-derived EVs were stained with two orange membrane dyes: ExoBrite™ 555/575 True EV Membrane Stain, and DiI. The stained EVs were diluted and run on a ZetaView® particle analyzer. Total EV number was measured in scatter mode, and fluorescently-stained EV number was measured with 520 nm excitation and 550 nm filter. The percentage of particles stained with the dye is shown in the plot. ExoBrite™ 555/575 True EV Membrane Stain showed nearly complete coverage of EVs, much higher than DiI.

 

Proven Results: ExoBrite™ True EV Membrane Stain Outperforms PKH67 for EV Detection on ARC™ Particle Analyzer

 ExoBrite™ 515/540 True EV Membrane Stain is more effective at staining EVs and is less susceptible to dye aggregation than PKH67 membrane dye.
Fluorescence analysis of HEK-293 cell-derived EVs stained with ExoBrite™ 515/540 True EV Membrane Stain and PKH67. The results indicate ExoBrite™ 515/540 True EV Membrane Stain is more effective at staining EVs than PKH67 membrane dye (62% vs. 49%, respectfully).
