Bright EV Stains with Excellent Signal-to-Noise

ExoBrite™ CTB EV Staining Kits are fluorescent conjugates of cholera toxin subunit B (CTB), optimised to provide bright, specific staining of extracellular vesicles (EVs) while overcoming common challenges in EV detection.

Unlike many lipophilic dyes that form aggregates or bind non-specifically to polystyrene beads, ExoBrite™ CTB stains are formulated to deliver high fluorescence intensity with little to no background aggregation. This makes them highly reliable for flow cytometry detection of both purified EVs and bead-bound EVs.

By binding to GM1 gangliosides commonly found on EV membranes, ExoBrite™ CTB stains ensure superior signal-to-noise ratio and more complete coverage compared to traditional stains such as DiO, CellMask™, ExoFlow-ONE™, and ExoGlow™. Fully compatible with antibody co-staining (e.g. CD9, CD63, CD81), these stains enable multi-parameter EV analysis with confidence.

Each kit contains ExoBrite™ CTB EV Stain and Reconstitution Solution, yielding 100 µL of 500X staining solution per vial.

ExoBrite™ CTB EV Stains deliver exceptional signal-to-noise and more complete coverage of purified and bead-bound EVs compared to other stains. In contrast, lipophilic dyes like DiO and CellMask™ show significant aggregation, while stains such as ExoFlow-ONE™ and ExoGlow™ provide less EV coverage.

ExoBrite™ CTB EV Stains show lower background and more complete EV staining than competitor dyes. EVs purified from MCF-7 supernatant (SEC) were stained with the indicated dyes (top row). Filtered PBS controls (bottom row) reveal dye aggregation, highlighted with red arrows. Lipophilic dyes like DiO and CellMask™ show high background, while ExoFlow-ONE™ and ExoGlow™ provide less-complete EV coverage compared to ExoBrite™ CTB.

Proven Performance

SEC-purified MCF-7-derived EVs were stained with ExoBrite™ CTB EV Stains.

ExoBrite™ 490/515 CTB EV Stain was used to stain either SEC-purified, Jurkat-derived EVs, or artificial liposomes. Robust staining was seen with the EVs, but the liposomes were not stained. The liposomes in this experiment were prepared from Presome® ACD-1, and were verified to be of similar size and concentration to the EVs using the lipophilic dye di-8-ANNEPS (data not shown). Analysis was performed on a CytoFLEX flow cytometer with SSC detected from the 405 nm laser, and ExoBrite™ detected in the FITC channel. Presome® is a registered trademark of NIPPON FINE CHEMICAL CO., LTD.

SEC-purified MCF-7-derived EVs were stained first with ExoBrite™ 490/515 CD9 in 100 uL, followed by 10X ExoBrite™ 560/585 CTB EV Stain. EVs were detected on a CytoFLEX LX flow cytometer in the FITC and R-PE channels. When we gated on ExoBrite™ 560/585 CTB-positive particles, ~95% were also positive for CD9.
