Next-Generation Tyramide Amplification Dyes for Brighter, More Stable Multiplex Spatial Biology

TyraMax™ Amplification Dyes are Biotium’s next-generation tyramide signal amplification (TSA) reagents, engineered to meet the growing demands of spatial biology and high-plex imaging. Compared to earlier CF® Dye tyramides and other TSA dyes on the market, TyraMax™ offers clear advantages in signal brightness, photostability, and working solution stability.

Designed for applications such as immunohistochemistry (IHC), immunocytochemistry (ICC), and in situ hybridization (FISH), TyraMax™ dyes deliver robust performance in both manual and automated staining workflows. Notably, TyraMax™ dyes remain stable in amplification buffer for at least 24 hours, including near-infrared dyes—an important advantage for high-throughput and automated platforms.

Power Your Multiplex Panels with TyraMax™

Tyramide signal amplification (TSA), also known as Catalyzed Reporter Deposition (CARD), is a highly sensitive method for detecting low-abundance targets. Its compatibility with cyclic immunofluorescence (CycIF) has made TSA indispensable for modern multiplex and spatial biology workflows.

TyraMax™ dyes were developed to push TSA performance further, delivering brighter and more photostable signals than Aluora® and Opal® reagents, while also offering improved chemical stability. This makes TyraMax™ particularly well suited for high-plex panel development, spectral imaging, and long acquisition times.

Biotium’s far-red/near-IR TyraMax® Amplification Dyes offer advantages in brightness and photostability compared to other TSA dyes. PFA-fixed HeLa cells stained with WGA-HRP detected with TyraMax™, Opal™ Polaris, or Aluora™ Spatial dye. Imaged using a 40X oil objective on Evident FV4000 confocal system with the indicated laser line. Photostability measurements were done on an Olympus IX71 epifluorescence microscope using a Cy®5 filter cube. Stained cells were imaged every 10 seconds for 120 seconds, and mean fluorescence was normalized to the first image taken.

TyraMax™ 740/770 has excellent brightness, photostabilty, and working solution stability, making it compatible with automated workflows. PFA-fixed HeLa cells stained with WGA-HRP detected with TyraMax™ 740/770 Amplification Dye or Aluora™ 750 Spatial dye and imaged in PBS using a 40X oil objective on Evident FV4000 confocal system with 730 nm laser in the Alexa Fluor® 700 detection channel. For photostability measurement, images were collected every second during continuous illumination at 10% laser power. Mean fluorescence at each time point was normalized to the fluorescence for the first image captured. For working solution stability testing, OD-matched solutions of TyraMax™ 740/770 and Aluora™ 750 dyes were prepared in Tyramide Amplification Buffer Plus with 0.0015% hydrogen peroxide. Absorbance was measured on an Agilent Cary 60 spectrophotometer, and then the solutions were left at room temperature, protected from light, and absorbance was measured at various time points up to 24 hours. TyraMax™ 740/770 was stable in the presence of hydrogen peroxide over 24 hours, while Aluora™ 750 degraded almost completely.
